Resumo: The release of drugs at the colon shows itself as a therapeutic alternative for pathologies that affect this organ, to the absorption of peptidics drugs and proteins and in chronopharmacotherapy. The systems intended to release the drugs at the colon must be formulated considering characteristics as pH, enzymatic activity, microflora and time to get in to the gastrointestinal tract. This research has by objective to present a drug delivery system directed to the colon, using natural polymers as zein and chitosan associated with ethylcellulose. These polymers were prepared in order to obtain polymers dispersion effective at application of tablets coating considering solubility, pH, viscosity and proportion among polymers. The therapeutic system proposed comprehended the use of zein as sealed layer isolating the center, which contains the drug, of the aqueous functional coating and, blend of ethylcellulose and chitosan as functional coating promoting the modified released. This system was evaluated for the coating process and in vitro dissolution profile of tablets containing the model drug propranolol hydrochloride. The polymer films obtained by casting were characterized by FTIR, thermal analyses, mechanic analyses and water vapor transmissions. The obtained results showed that zein dispersed in hydroalcoholic solution can be used as film and coating layer, in coating system, isolating the center of incompatibles components and, as alternative for the protection of moisture sensitive drugs, acting as barrier. This characteristic is due, in grand part, to the presence of apolar groups and water insolubility. The chitosan is one polysaccharide specifically degraded by the colon flora and presents swelling characteristics and film forming, being useful as specific delivery in the colon. Presented applicability when it is used in combination with ethylcellulose, where the ratio of polymers and the coating load influenced directly the profile of drug dissolution. The association of polymers with different physicochemical characteristics makes the release of the drug a complex mechanism, based on the swelling of the coating film by the gastrointestinal tract and system eruption. The result of the thermal analyzes and FTIR are indicatives that there was no incompatibilities among the used materials and, the mechanic analyzes identified bigger intermolecular interaction in the polymeric blend. It is concluded that zein and chitosan biopolymers presented desirable characteristics to the application of tablets coating, also biodegrability, biocompatibility and renewable source, being viable your application in a modified release system.
