Peptic ulcer disease, which includes gastric and duodenal ulcers, is a chronic disease with multifactorial etiology that affects about 10% of the world population. The biggest cause of the appearance of ulcers occur by endogenous agents (increased secretion of acid and pepsin) or exogenous (Helicobacter pylori, NSAIDs, ethanol, stress, ischemic processes, among others). The treatment of peptic ulcers is made by the use of drugs proton pump inhibitors (such as omeprazole and lansoprazole) and histamine receptor blockers (such as cimetidine and ranitidine). However, these drugs have harmful side effects and are not able to promote the healing of gastric mucosa efficiently, which usually causes disease relapse. The search for new substances for treating this disease involves the use of natural products. Solanum paniculatum L., target species of this study, is cited for antiulcerogenic properties and is present in RENISUS, however, there are few studies that elucidate their pharmacological properties against this disease. Therefore, the aim of this study was to evaluate the gastroprotective and healing effect of the hydroalcoholic extract of the leaves of S. paniculatum (EHSPFl) against the experimental peptic ulcers. Was observed that EHSPFl presents significant gastroprotective effect against ethanol and indomethacin (NSAIDs) damaging agents and the acute ischemia and reperfusion, with the effective dose of 62.5mg/Kg. The extract acts in gastroprotection by both local and systemic action, being able to reduce the levels of LPO, increased mucus production and reduce neutrophil infiltration, in addition to acting via modulation of NO. However, the extract was unable to restore the levels of GSH, SOD and CAT in acute models, and did not decreased acid secretion. In the curative model of gastric ulcers induced by ischemia and reperfusion, we observed effective protection of EHSPFl with reduced LPO and MPO, increased ... |
