Influência do resveratrol sobre metabolismo energético, estresse oxidativo e expressão SIRT1 no miocárdio de ratos diabéticos

Resumo: Studies suggest Diabetes mellitus (DM) promotes alterations on energy metabolism and oxidative stress, which provide important factors to initiate cardiovascular disorders. The use of antioxidants has aroused interest on clinical due therapeutics properties on cardiovascular diseases, and may thus prevent morbidmortality in diabetic patients. In this sense, it is proposed to study the effect of resveratrol (RSV - 3,5,4 '- trihydroxystilbene) via serum biochemical parameters, energy metabolism and oxidative stress in heart tissue of rats with diabetes mellitus type 1 . 32 rats, Wistar race, with ± 250g body weight were used. The animals were divided into experimental groups (n = 8) : G1 (normal control), G2 (normal, treated with resveratrol), G3 (diabetic untreated) , G4 (diabetic treated with resveratrol) . The experimental Diabetes mellitus was induced by the intraperitoneal administration of streptozotocin (STZ) in a single dose, at a concentration of 60 mg / kg body weight. The animals in Groups G2 and G4 received RSV via gavage at 1.0 mg/kg/day for 30 days. At the end of the experimental period (30 days), nutritional parameters were evaluated and determined to glucose, triglycerides (TG), total cholesterol (TC), lipoprotein fractions and lactate dehydrogenase (LDH) activity. In cardiac tissue, concentration of total protein, glycogen, lipid hydroperoxide (HP), total glutathione, reduced glutathione, glutathione peroxidase (GSH-Px) and superoxide dismutase (SOD) was determined and activity of marker enzymes of energy metabolism, lactate dehydrogenase (LDH), citrate synthase (CS) and β-hydroxyacyl hydroxylase (OHADH), and the expression of situín protein (SIRT1) through Western Blotingt.
