Bioestimulação de célula-tronco mesenquimal de rato sob ação da luz contínua e pulsátil de 630 nm utilizando LED

Resumo: Regenerative medicine (RM) has emerged of different sciences boost, including biology of the stem cell. The mesenchymal stem cells (MSCs) isolated from adipose tissue have occupied more space in MR. They are characterized by differentiation, self-renewal and the ability to generate at least three different cell lines, being involved in the repair of damaged tissue, however they are dose-dependent requiring expansion by cell culture method. The applicability of LED as biostimulator agent has been proposed. The aim of this study was to evaluate the effect of continuous and pulsed light, 630nm, comparing this action in cells obtained by the methods of mechanical and enzymatic dissociation. Mouse fat blocks were removed from rats with an average age of 3 months. The concentration of cells obtained LMN / g tissue 1 x 105 and 3.34 x 105 MD and ED, respectively. The phenotypic profile using markers as CD90 / CD44 / CD71 / CD31 / CD11b / CD45 showed that ED removes a significant percentage of the surface antigens of MSCs. The curve of the confluence at 80% between passages showed that MSCs obtained by MD was beneficiated by the action LED and proliferating faster. Cytokines TNF, IFN-ɣ, IL-4 and IL-10 were determined by flow cytometry. Most of the samples showed cytokine secretion was obtained by MD method. Control samples, not-irradiated also showed secretion of cytokines, showing the action of the additive of the culture medium as biostimulator of paracrine effects. The micronucleus and comet assays to the genotoxicity analysis show that pulsed light of 1J / cm2 determine the appearance of micronuclei, while DNA damage determined by the comet assay decreases by the LED action
