Resumo: | Objective:To Identify Biological Markers Associated To Proteolytic Processes In Melanoma. To Perform Qualitative And Quantitative Analysis Of The Secretome Of Tumoral Cells And Patient Samples, As Well As To Identify The Main Post-Translational Modifications At The N-Terminus Of Secreted Protein, Correlating The Results With Proteolytic Processing Events In The Light Of Biological Pathways Related To Oncogenesis. Methods: We Performed A Proteomic Analysis Of Secreted Proteins From Four Human Cell Lines: (I) A Paired Set Of Fibroblasts - Hs 895. T, A Cell Line Obtained From A Lung Metastatic Site From A Patient Who Had With Melanoma And Hs 895. Sk, A Cell Line Of Skin Fibroblasts (Derived From The Same Patient); (Ii) Two Malignant Melanoma Lines - A375, A Malignant Melanoma Cell Of Primary Source Lineage, And Sh4, A Cell Line Derived From A Patient With Pleural Metastatic Melanoma. Terminal Amine Isotopic Labeling Of Substrates (Tails) Analysis Was Performed On Human Cell Lines (Hs 895 Sk, Hs 895 T, A375 And S |
