Resumo: Objective: To evaluate the macro and microscopic alkaline ocular chemical burn in rabbits, using amniotic membrane graft (MA), fixed with fibrin glue or sutures, maintaining epithelial layer superiorly or inferiorly face. Methods: Forty-five New Zealand rabbits lineage underwent anesthesia, the animals were randomized into five groups, with nine rabbits were burning standardized; nine animals with burn standardized and MA fixed with fibrin adhesive, keeping the face epithelial superiorly; nine animals with burn standardized and MA fixed with fibrin glue keeping the epithelial face inferiorly; nine animals with burn standardized and MA fixed with suture polyglactin maintaining epithelial face superiorly; and nine animals with burn standardized and MA fixed with polyglactin suture, keeping the face epithelial inferiorly. In the 30th day there was a performed macroscopically and histological evaluation. Statistical tests, depending on the nature of the variables, consider the statistical significance of p <0.05. Results: The analysis criterion in macroscopic adhesion showed no significant difference in the positioning and vascularization, but clinical appearance there was a slight advantage of the adhesive, but without statistical significance. The histological criteria in the epithelium, stroma, Descemet's membrane, endothelium, vessels, fibrosis, goblet cells, granulomas, retro corneal membrane, there were no significant differences, on the order hand inflammation and keratocyte there were differences, but not significant. Conclusions: The amniotic membrane graft in ocular chemical burns showed slight favorable results with the use of fibrin adhesive, as compared to suturing. Regarding the use of amniotic membrane epithelial superiorly or inferiorly face there was no significant differences.
