Resumo: Introduction: the use of gloves is closely linked to health care, it is one of the main ways to prevent Health Care Related Infections (HAI). It has two objectives, in which the first is the safety of the health professional and also the safety of the patient. The gloves make up the set of Personal Protective Equipment (PPE), where universal precautions have been introduced since the discovery of the Acquired Immunodeficiency Syndrome (AIDS) in the 1980s, now known as standard precautions. Objectives: Check the knowledge and use of sterile and non-sterile procedure gloves among nursing professionals. Method: This is a descriptive, cross-sectional, observational, analytical study with a quantitative approach. Data were collected through a questionnaire and a form. It was carried out in municipal and regional hospitals in the microregion of Alto Taquari. It was developed in 2 phases: 1. Application of two questionnaires, with the purpose of socio-professional characterizing the participants, and the other to assess the professionals' knowledge about the use of gloves during the procedures; 2. Non-participatory observation, using a checklist; Data were transferred to Software R, version 4.0.1.and the tests used were assisted by a statistician with experience in studies in the field of nursing. The project met all ethical precepts in accordance with the Resolution of the National Health Council (CNS) 466/12, CAAE 29990320.0.0000.0021. Results: A total of 87 research participants, who had the opportunity to observe 53 nursing professionals who worked in the aforementioned hospital units participated in the study. Professionals who participated in the survey expressed their views on the use of gloves in different activities. A wide range of opinions regarding the technical dressing procedure was observed. Chronic wounds with the use of instruments stand out, where no professional has the knowledge about the need to use gloves. Regarding the administration of medications, most PE claim to be correct to use LPNE to perform these procedures: intramuscular (91.8%), intradermal (89.5%), oral (80.2%) and subcutaneous (89.5%). Conclusion: it is concluded that nursing professionals do not have a good knowledge of the use of gloves, and this is reflected during the performance of some procedures, there are also flaws in practice. The importance of specific courses and training on the use of gloves during evidence-based care practice was observed. Keywords: Gloves; Infection; Health Care; Nursing Assessment.
