Dinâmica da formação de componentes da matriz conjuntiva extracelular em torno de materiais retrobturadores implantados em tecido subcutâneo de ratos

Resumo: The objective of this research was to study the changes in the components of ECM around two retro filling materials, MTA and Consistent Sealapex (SC), implanted in the subcutaneous tissue of rats, researching the expression, by indirect immunofluorescence (IFI), the presence of fibronectin, laminin, collagen, type III and type I. It was also evaluated the histological reaction and the ability of mineralization of these materials through the staining techniques of H&E, PIFG and Von Kossa. 24 rats were distributed in two protocols: For the study of collagens and glycoproteins, using IFI, six rats were placed in groups of three animals for each experimental material, SC, MTA and control during the periods of seven and 90 days. The histological protocol was done in 18 rats, divided in groups of six animals sacrificed in seven, 60 and 90 days of experiment. The response of the connective tissue in contact with the materials was evaluated in a descriptive and semi-quantitative ways and through the scores ( 0 = absent; 1= discreet; 2= moderate; 3 = intense), analyzing the inflammation, granulation tissue, fibrosis and calcification. The comparison between the different periods of study, the MTA group showed greater inflammation in seven days than in 60 days (p=0.0360), the Consistently Sealapex groups showed more granulation tissue between 7 and 60 days (p=0.0321). As the calcification, in all experimental groups it was observed positive granulations Von Kossa in all experimental periods. In the Group Consistent Sealapex calcification was more evident in 60 days when compared to seven days (p= 0.0217). Statistical differences were not observed in immunolabeling of glycoproteins and collagens, when compared between groups and between the periods of observation. After analysis of the results we conclude that, during the experimental time, the materials caused the same kind of histological reaction, both promoved calcification and fibrosis, there was a reduction of expression of fibronectin, increase of laminin, collagen type III and type I.
