During the covid-19 pandemic, much was said about possible treatments and prevention mechanisms for the disease, before vaccines and more recent studies on effective drugs for its cure emerged. Among them the chloroquine the indication became controversial, due to possible side effects and also due to the uncertainty of its efficacy. The media have been blamed for the excessive politicization of the use of the product, particularly in the first year of the pandemic. This study seeks to understand how citizens were influenced about the use of chloroquine, particularly by the written media, from April to June, 2020, considering the speech of actors such as the President of Brazil, Governors, Professors, Health Minister, Health Professionals, Consultants and World Health Organization ─ WHO, in articles published in two major newspapers in the state of São Paulo: O Estado de S.Paulo and Folha de S.Paulo. The theoretical referential on Discourse Analysis, Content Analysis and Semiotics was followed by the use of the web scraping technique, which resulted in the obtainment and analysis of 539 reports mentioning the use of Chloroquine in the treatment of covid-19. This content analysis of the reports was followed by the application of a questionnaire to 130 mentioned newspapers readers (printed or online version), in order to understand their perception regarding the actors speech identified in the collected articles. Finally, a semantic analysis of five of the collected articles was carried out, using as a script the concepts of semiotics and Content Analysis, proposed by Bardin ([1977] 2011). Through said methodology, with qualitative research, it was possible to understand that, although there has been space for the different actors in the mentioned newspapers, their speech, in general, was little relevant and insufficient to influence the researched sample. In this regard, the actor President of Brazil was the most cited in the analyzed articles; on the other hand, in the perception of the respondent readers, his speech was unable to influence them. A caveat should be made to the speech of the actors Doctors and World Health Organization ─ WHO, which although less cited in the reports than the President of Brazil were indicated by respondents as capable of greater influence on their opinion formation regarding the use of chloroquine in the treatment of covid-19.
