Senescence is a cellular response to severe stress and damage, characterized by cell cycle arrest, upregulation of pro-survival pathways and induction of a complex secretory phenotype. Cellular senescence is an age-related process, being argued that the percentage of senescent cells increases with age. Clearance of senescent cells has proven efficiency in reducing senescent effects in several in vivo models, and the use of senolytics as a therapy seems a promising tool to fight aging and age-related diseases. Recent research has identified targetable vulnerabilities of senescent cells that can be exploited by a novel group of drugs, senolytics. Despite successful proofs-of-concept using senolytics, their potential translational application is hampered by their associated toxicities. Here, we created nanoparticles able to release senotherapeutics and eliminate senescent cells. The senolytic activity of the nanoparticles was obtained by high-throughput screening against healthy and senescent cells.
