The cajazeira (Spondias mombin L.) is distinguished by producing fruit of excellent taste and nutritional properties. Nevertheless, there are no commercial crops and places of occurrences of occurrence are suffering in deletions due to human action. This study aimed to evaluate the genetic diversity among plants Cajazeiras Bank of germplasm of the IPA by ISSR markers. The experiment was conducted at the Laboratório de Plant Biotecnologia Vegetal, Department of Agronomy, of the Federal Rural University of Pernambuco (UFRPE), in Recife-PE, with latitude of 8 10'52''S and longitude 34 ° 54'47'' W, from July 2007 to May 2009. Tender leaves were collected from twelve plants (Charuto, Djalma, Vianey-1, Vianey-2, Vianey-3, EAN-1, EAN-2, Casa Grande, Muribeca, IEE (Estação Experimental de Itapirema), Dr. Moacir e Limão) from Germplasm Bank of the Cajazeiras at the Experimental Station of the Agronomic Institute of Itambé Pernambuco (IPA) (7 ° 24'50 "S and 35 º 06'30" W) in the Municipality of Itambé, Pernambuco, Brazil. Genomic DNA was obtained according to the methodology proposed by Ferreira and Grattapaglia (1998), with modifications.For the amplification of DNA, 18 primers were used (2 UBC, UBC 808, UBC 810, UBC 812, UBC 813, UBC 820, UBC 827, UBC 834, UBC 849, UBC 855, UBC 860, UBC 866, UBC 868, UBC 881, UBC 884, UBC 885, UBC 887, UBC 888 and UBC 891). These amplifications generated 135 fragments, of which 105 polymorphic, ranging from 20 to 620pb. The UBC 834 was more informative, allowing the differentiation between the 12 accessions. The primers that prevails in the sequence of bases GA (2 UBC, UBC 810, UBC 812 and UBC 885) the highest number of amplified fragments and polymorphic fragments. In the construction of the dendrogram was the formation of four distinct groups, from an average similarity of 51.69%. Access Dr. Moacir, comes from Manaus-AM, was isolated from the other accessions, presenting a similarity of 27.45% with access EAN-1, the lowest found among the 12 accessions. Already between 1 and Vianey Vianey 2 there was a greater similarity among all accessions (73.21%). With these data we can see the viability of the use of ISSR markers to study genetic diversity in germplasm of Cajazeiras and high geneticdiversity among accessions of the bank. |
