The use of molecular biology in animal reproduction to identifying the sex genetic allows the producer has better reproductive planning, facilitating the coordination of actions aimed at rationalizing production and profitability. The objective of this study was to determine methods for the identification of the sex of the goats and sheep, using different concentrations of DNA (150, 100, 50, 1, 0.5, 0.2 and 0.1 ng) extracted from blood of both sexes, as a possibility for application in sexing embryos pre-implantating, sperm and animals pseudo-hermaphrodites. It was performed polymerase chain reaction (PCR) multiplex for the SRY gene (116bp) found in the Y chromosome, the gene for Aml-X (300pb) found on the X chromosome, in addition the sequences corresponding to genes ZFX / ZFY containing 445 and 447bp respectively. The use of these methods presented showed accuracy of 100% in samples of blood goat and sheep, in low concentrations, at least 0.2ng (SRY/Aml-X) and 1.0ng (SRY/ZFXZFY), which suggests the feasibility of the technique for determining sexual in these species. |
