Studies on the development of adipocytes have become important due to the fact that differentiation of this cell type is associated with several chronic and or degenerative diseases such as hypertension, diabetes, cardiovascular disease and cancer, which have link with obesity due in part to the expansion of adipose tissue. Vitamin D has shown relationship with obesity and their molecular bases, pathways and interaction with tissues, such as adipose tissue, therewise studies are necessary to support future clinical practices and actions to control of diseases that affecs large populations. This research aims to study these interactions in vitro using SGBS human adipocytes to assess the expression of VDR, GR and PPARɣ genes. For this study, three independent biological replicates were made separate at treatment times (4 hours, 12 hours, 24 hours, 4 days , 8 days and 12 days) and pre- adipocyte cells from time 0 hour. At all times the cells were evaluated divided into two groups: control (ethanol) and treated (1,25(OH)2D3). Total RNA extraction with subsequent analysis of gene expression by RT-qPCR were performed. When comparing the relative expression of target genes with untreated pre-adipocyte (0h) for both groups and times, the GR gene showed a statistically significant difference ( p < 0.001 ) for the group treated with vitamin D in 12 hours, 24 hours, and, 4, 8 and 12 days for the control group and the times 12 and 24 hours, 4 and 8 days. There was significant difference for PPARɣ gene in the group treated at times 8 and 12 days, similar to the control group results, when compared with the pre-adipocyte (0h). The basal expression of the VDR gene showed no statistically significant difference when compared with the preadipocyte. However, for the fold change, the VDR gene was the only to show expression statistically different at time 0h having a negative expression of 2.87 (p <0.05) at time 4 days. The values of fold change found in... |
