Resumo: | The Haemonchus contortus stands out as the main worms in sheep. This seems to be more likely to the development of resistance to antihelmintics, probably due to its high biotic potencial and genetic variability. The cryopreservation allows lineage maintenance with desirable characteristics for a long period of time, added to a drastic decrease in the number of animals kept in laboratories. The present study aimed at comparing different cryopreservation techniques, considering the best results regarding survival rate, motility and infectivity maintenance of Haemonchus contortus L3 larvae. The difference between each technique occurred through the alternation between three distinct stages, the conservation or removal of the sheaths of Haemonchus contortus L3 larvae with Sodium Hypochlorite, the use or not of Ethylene Glycol as a cryoprotector and the freezing speed, slow or ultrafast. The study identified strategic stages on the cryopreservation of Haemonchus contortus L3 larvae, among these, the absence of sheath and slow freezing proved to be essential for obtaining any survival rate after the defrosting. Instead, the use of Ethylene Glycol as cryoprotector was not indispensable, however presented technical benefits, significantly increasing the survival rates and motility for a longer period of freezing |
