Avaliação das modificações de historias H3Ki36me3, H3K9ac, H4K1 2ac e H3SI0ph em tumores odontogênicos em diferenciação ameloblástica

Resumo: | Odontogenic tumors are a heterogeneous group of lesions with varied biological behavior. Epigenetic modifications have earned attention in the study of carcinogenesis as being among the most common abnormalities in human neoplasms. Histone modifications are epigenetic modifications intimately related to the relation with the regulation of the cell cycle and direct relationship with the process of tumorigenesis. The aim of the present study was to investigate the expression of antigens related to histone modifications in samples of benign and malignant odontogenic tumors with ameloblastic differentiation using immunohistochemical assay. To this end, the cases assessed included 8 ameloblastomas, 4 adenomatoid odontogenic tumors, 6 calcifying cystic odontogenic tumors, 5 calcifying epithelial odontogenic tumors, 4 adenoid ameloblastomas, 12 amelobastic carcinomas, 6 clear cell odontogenic carcinomas and 3 ghost cell odontogenic carcinoma. After histopathological review of the cases, immunohistochemical reactions were performed with antibodies anti-histones H3K36me3, H3K9ac, H4K12ac and H3S10ph. Immunoreactivity and staining area were assessed for each antibody in all tumors and between benign and malignant groups. These parameters were evaluated with the software ImageJ. Immunoreactivity and staining area were statistically different for H3K36me3, H3K9ac and H4K12ac between benign and malignant groups. Statistical difference was also found in the immunoreactivity assessment for H4K12ac among the tumors. With respect to the staining area, there was statistical significance for H3K36me3 and H4K12ac among the tumors. It could be concluded that the variation of histone modifications is related to carcinogenesis. |
