In Brazil, the discussion about the change in the energy matrix is gaining a bigger dimension. In this context appear the interest in studying plant species such as physic nut (Jatropha curcas L.), because it is an oilseed crop that has major socio economic potential for the biodiesel production. This way, arises the need to produce large quantity of seedlings selected from superior mother plants. The aim of this study was to evaluate the influence of growth regulators on in vitro establishment and callus induction in physic nut (Jatropha curcas L.). Leaf segments of selected mother plants of accession JCUFS-012 of the active germplasm bank of the UFS were used as explant source. The plants were grown in a greenhouse with white screen. In this work we evaluated the effect of sodium hypochlorite concentrations, time of immersion of explants types and concentrations of the plant regulators BAP, IBA, NAA, 2,4-D, IAA and kinetin on in vitro establishment, callus induction and anatomy of the tissues during organogenesis of physic nut. For in vitro establishment and regeneration we recommend the use of 0.5% of sodium hypochlorite for 5 minutes for the disinfestation of leaf explants of physic nut. Explants from leaf number 3 (the third leaf from the apex to the base, 9.4 cm wide and 10.0 cm length), are the most effective for callus induction. The use of MS medium supplemented with BAP (0.5-1.0 mg L-1) and IBA (0.5 mg.L-1) is essential for induction of compact callus in physic nut. Histological analysis showed formation of developed adventitious buds, with cambium connection with the callus tissues, evidencing indirect organogenesis. |
