Citotoxicidade de medicações intracanais em fibroblastos L929

Resumo: Intracanal dressings are substances used during endodontic treatment and requiring biocompatible with periradicular tissues, not causing tissue damage or interfere in repair process. This study aimed to assess the cytotoxicity of endodontic dressing when in contact with L929 mouse fibroblast cells, after different periods. To assess the cytotoxicity of three intracanal dressings and three vehicles were divided into seven groups, namely: calcium hydroxide/camphorated paramonochlorophenol/ glycerin (CPG); iodoform/ glycerin (IG); calcium hydroxide/iodoform/distilled water (CIW); iodoform/distilled water (IW); calcium hydroxide/distilled water (CW); Otosporin® (OT); and control group. The eluates of the medications were prepared and placed in contact with the cells (1 x 105 cells/well) during periods of 24, 48, 72hours, 5 and 7 days. After each evaluation time, a colorimetric assay was conducted using methyl tetrazolium reagent (MTT) and the reading of the plates was performed in a spectrophotometer with optical density of 570 nm. Data were tabulated and subjected to statistical analysis by ANOVA-Tukey test with a significance level of 5%. At 24hours, IG and OT (P <0.001) showed greater cytotoxicity, as also observed by the OT group (P <0.001) when analyzed in 48hours. In 72 hours, it could be observed that the CW and OT groups were more cytotoxic compared to the control group. These data were also demonstrated in groups CPG, IW, CW and OT in 5 days. Within 7 days all drugs showed cytotoxicity, but OT group showed the most cytotoxic (P <0.001). With respect to experimental time factor was observed in all groups significant differences between 24hours and 7 days. It is concluded that all medications showed cytotoxicity at some point in the research being Otosporin ® the most cytotoxic and that time influenced the cytotoxicity of all medications analyzed intracanal medication.
