Mushrooms are known for medicinal value and it has health benefits associated with its dietary intake, mainly Agaricus bisporus due to its various bioactive components. However, there are difficulties to determine bioactive compounds, since the different extraction methods can result in variate chemical profile and frequently they are not investigated, as well as the toxicity of residual solvents are not considered. The supercritical fluid extraction (SFE) uses high-pressure of CO2 for its application as solvent; it has considered a clean technology, since the extracts obtained with this process have high purity with low residual solvents. We evaluated different extracts from Agaricus bisporus using supercritical fluid extraction (SFE) and classical organic solvent extraction (crude extract), with ethanol as solvent, comparing their yields, chemical profiles, cytotoxic, antiaggregant and anticoagulant activities. The chemical profile was evaluated by Gas or Liquid chromatography coupled to mass spectrometry. The chemical analysis showed that methyl (E,E)-9,12- octadecadienoate and methyl (Z)-9-octadecenoate were the main esters derived from fatty acids in SEF extracts. The SFE had a high cytotoxic effect in the MCF-7 cell line, when compared to the MDA-MB-231 line. The McCoy cell line (non-tumor) showed no cytotoxic effect. Additionally, the anti-aggregate effect of SFE extracts was higher than the effect produced by classical organic solvent extraction. Overall, SFE extracts showed the best performance on bioactive screening compared to conventional ethanolic extract. |
