Estudo clínico-andrológico e genético-molecular da subfertilidade em touros da raça Gir Leiteiro (Bos taurus indicus) no Estado de Minas Gerais

Resumo: | The Dairy Gyr breed has been the subject of great interest because of its adaptation to climatic conditions in Brazil and the highly expressive milk production, but with reproductive difficulties reported. This study aimed to characterize the frequency of animals with testicular hypoplasia (TH) , the correlation between the number of copies of the testis-specific gene protein Y-ecoded (TSPY) between normal bulls (N) and TH , and to identify candidate genes for sub-fertility with bases in genome wide association study (GWAS) in Dairy Gyr bulls . 311 bulls from six farms in Minas Gerais (with and without nutritional supplementation), underwent soundness examination. Age, body weight (BW), scrotal biometry, physical and morphological semen analyzes were compared. Qualitative variables were analyzed by Mann-Whitney test and quantitative by Student t-test (P<0.05). 12 animals with TH and 12 N were selected for the analysis of copy number of TSPY. The amount of these was established by real time PCR and compared by Student t-test (P<0.05). GWAS for 60 individuals (30 affected and 30 N) were genotyped. These individuals were genotyped with the Affymetrix platform Axion Genome-wide Bos 1 array, 648k. The phenotype mapping is used in the residue of the effects of age, supplementation , date of collection and sire. The results were filtered to exclude SNPs with low quality results MAF or less than 0.03 and considered significant when - log10 (P value) 5. It was observed that the TH occurred in 5.79% of bulls. It was also observed in the parameters studied recorded difference (P<0.05) between bulls N and HT, except BW. The number of copies of TSPY was different (P<0.05) between N (164.6) and TH (172.5) bulls. There were associations with sub-fertility and genes on chromosomes BTA11, BTA16 and BTAX. It was conclude that there were TH bulls in herds that independent of nutrition. The number of copies of TSPY TH animals was higher than N ones. And there were associations between genes located on chromosomes BTA11, BTA16 and BTAX and sub-fertility in Dairy Gyr |
