Resumo: | The conservation of FIla Brasileiro dogs depends on projects and actions related to genetic characterization of the main in vivo preserved bloodlines and its comparative analyses, the creation and maintenance of banks for the in vitro preservation and an educational work directed to the association of conservationist breeders. The aim of the present study was to genetically characterize a pure Fila population and to test different extenders for the semen freezing process for the breed. Two experiments were conducted. In the first experiment 40 Fila Brasileiro. phenotipically cassified according to Paulo Santos Cruz standart, were genotyped using 21 DNA microsatellites marker (ISAG, 2008). In the second, clinical andrological examinations and semen collections of six Fila were perfomed. Characteristics of semen were evaluated immediately after sampling and also its freezability in four different extenders. Motility, as well as posthawing characteristics, were obtained by CASA and by Functional tests for the integrity of spermatic membranes (HOST, FCDA and PI) during a thermo-resistance test. Successful semen cryopreservation were obtained and allows the artificial insemination (AI) with frozen stored semen usefull for preservation programs and breeding. It can be concluded tha genotyping Fila Brasileira-PSC dogs using genetic analyses from DNA microstallite markers can help Fila preservation programs and breeding, specially associated with the use of frozen semen AI. |
