Influência do varespladib (ly315920) sobre o efeito paralisante da crotoxina em preparações neuromusculares de camundongos

Resumo: | According to the World Health Organization, approximately 5.4 million snake bites occur each year, with 2.7 million poisonings that lead to 100,000 deaths. In Brazil, one of the snakes of greatest medical importance is Crotalus durissus terrificus, which causes deaths in 1.1% of poisoning cases. The crotalic venom has high concentrations of phospholipase A2, one of which is the basic component (CB) of Crotoxin (CTX). This toxin is responsible for neurotoxicity and myotoxicity in crotalic accidents. Recently, Varespladip (LY315920), a synthetic phospholipase A2 inhibitor, has been studied to neutralize snake toxins. Therefore, to evaluate the inhibitor potential of Varespladib against Crotoxin, myographic and morphological experiments were performed, using mouse phrenic nerve-diaphragm muscle preparations in vitro. The preparations were exposed to CTX, CB, Varespladib or the pre-incubated product of CTX-Varespladib and CB-Varespladib. The results of the present study demonstrated that Varespladib is able to neutralize the paralyzing and myotoxic effects of CTX and the paralyzing effects of CB in mice neuromuscular preparations. |
