The Curcumin is a polyphenol that has many biological activities, however, has low absorption, thus resulting in reduced bioavailability. Soon the objective of this study is to biotransform curcumin in a potentially antioxidant product and more water-soluble. To do this, it is used the enzyme apparatus filamentous fungi, which make possible perform several structural changes in a single reaction step, thereby facilitating the application of the principles of green chemistry. So, two strains were selected from Beauveria (Beaveria bassiana ATCC 7159 and Beauveria of Cerrado IP 94) for the biotransformation of curcumin. Both were incubated in liquid glucose medium and had reactions monitored by Thin Layer Chromatography (TLC) and High Performance Liquid Chromatography (HPLC) for 168 hours. The biotransformation by both strains gave similar products, however only the Beauveria of Cerrado IP 94, was able to provide a satisfactory yield of the derivative, to perform of the complete characterization. Characterization of the derivative was proposed by mass spectrometric analysis and nuclear magnetic resonance (¹H NMR, 13C NMR, HSQC e HMBC), yielding reduced and metilglicosilado derivative, called LaBiocon 552. In an attempt to optimize the biotransformation, were evaluated two more culture mediums (Czapek and PDSM) with Beauveria of Cerrado IP 94. The PDSM medium shown to be capable of providing a higher concentration of derivatives, however the Czapek medium did not show the formation of any product. The antioxidant activity was evaluated by voltammetry method, and the derivative presented a satisfactory antioxidant activity, though lower than that of curcumin. However, when assessed solubility, derived pointed be approximately 9 times more water soluble curcumin. Evaluation of solubility was carried out by UV-Vis spectrophotometry. |
