Urinary disorders such as detrusor-sphincter dyssynergia are the main sequelae of acute spinal cord injury cranial to the sacral spinal segments. Therefore, studies have been performed with a focus on understanding the responses of the urinary bladder injured due to detrusor-sphincter dyssynergia and in the development of efficient therapies. Thus, the present study aimed to evaluate the urinary bladder histomorphology from rats submitted to a detrusor-sphincter dyssynergia model and treated with dantrolene and mesenchymal stem cells, as well as to quantify smooth muscle, elastic fibers and type I and III collagens after 30 days of spinal cord injury induction. The urinary bladders were collected from rats that were divided into: control group G N (n=5), trauma group G T (n=5), dantrolene group G DAN (n=5), mesenchymal stem cell group G CM (n=5) and association group G CMD (n=5). The urinary bladder samples were stained with hematoxylin and eosin, Weigert-Van Gieson and Picrosirius Red. Immunohistochemistry was performed to monitor the migration of genetically modified mesechymal stem cells for expression of green fluorescent protein to the injured bladder. The mesenchymal stem cells, but not dantrolene or association, maintain the amount of elastic fibers and type I and III collagen fibers to the urinary bladder of Wistar rats 30 days after trauma. Besides that the mesenchymal stem cells migrates to the urinary bladder after lesions consequent of detrusor-sphincter dyssynergia induced by experimental suprassacral spinal cord injury in Wistar rats. |
