Estabelecimento de modelo para prospecção de fármacos antiangiogênicos na membrana do saco vitelino de embrião de galinha

Resumo: Angiogenesis is directly linked to molecular processes including endothelial growth to develop new vessels through production of physiological and pathological factors, but the mechanism involved in stimulation and inhibition of such process is not yet fully elucidated. Therefore, the objective of this work is to establish a model for prospection of antiangiogenic drugs in chicken embryo yolk sac membrane. The study was divided into two phases, first were used fertile chicken eggs packed in an electric brooder for 72 hours, at 60h time a 5 mL albumen withdrawal was performed, then the eggs were returned to the brooder. Within 72 hours of the first phase, an wide opening of the shell of each egg was carefully performed, and during this opening an additional 15 mL of albumen was withdrawn. After opening, the test substances (saline and bevacizumab) were inoculated in two different ways, retained (focal) and non-retained (non-focal) to a given field. After this, the second phase was started, where the eggs were kept in a CO2 incubator with a controlled temperature of 37 ° C for a further 48h. During this time period a 12-hour time interval follow-up with 4X increase picture was recorded for non-focal method and 10X for focal method. For image analysis, manual and automatic methods were used. The study results were obtained with two evaluation methods (manual and automatic) in two approaches (focal and non focal). It was observed that the vascular development profile presented in all measures significantly higher in the control group (inoculated with saline solution) compared to the Bevacizumab group in both models, showing the antiangiogenic action of the drug. When Pearson's linear correlation coefficient was performed between the manual parameters si and the crossing between the manual and automatic parameters, the correlations were extremely positive. It was possible to conclude that angiogenesis model in the yolk sac membrane, in both focal and non-focal approach, provides temporal monitoring of angiogenic process and enables the demonstration of drugs antiangiogenic effect.
