Resumo: |
Cryptococcosis is an opportunistic systemic mycosis caused mainly by inhalation of fungal propagules of the species Cryptococcus gattii and C. neoformans in its two varieties, var. grubbiand var. neoformans, whose pathogenicity is based on the expression of virulence factors directly involved in the establishment and maintenance of infection, and is related to the production of capsule, melanin, lipases, proteases and urease, in addition to biofilm formation. The antifungal drugs available for the treatment of cryptococcosis is currently limited and restricted to a few classes of drugs, which generates the need to carry out research studies of new antifungal drugs and the discovery of new molecular targets in Cryptococcus. In recent years, several studies have shown the potential antifungal the drug inhibiting synthesis of folic acid in pathogenic fungi, and before that, we propose to investigate in vitro effects of folate inhibitors in C. gattii and C. neoformans, evaluating the action of these drugs on the sensitivity, antifungal resistance and production of virulence factors. To this end, we analyzed the antifungal susceptibility profile of strains of C. gattii (n=15) and C. neoformans (n=15) compared the combination of SMT-TMP and SDZ-PIR, and profile of resistance to antifungal drugs. Additionally, we investigated the effect of folate inhibitors on the production of virulence factors and biofilm formation in strains of C. gattii (n=7) and C. neoformans (n=7) studied. Was collected from in order to investigate possible environmental niches preferred strains of Cryptococcus in the city and metropolitan region of Fortaleza, Ceará. All strains tested were sensitive to the inhibitory action of the drugs tested, but showed no significant effects on the virulence factors, except for biofilm formation. Thus, we believe that the results of this study may contribute to greater understanding of the antifungal potential of folate inhibitors on species of Cryptococcu |
