Estudo da correlação entre a imuno-expressão de tnf- α, nf-кb, cox-2, e ki67 com fatores prognósticos em carcinomas orais

Resumo: Cancer is a disease of great concern due to its high incidence and high mortality and is currently considered the second largest cause of death in the world. In the oral cavity, squamous cell carcinoma is the most prevalent malignant neoplasm. The treatment of this tumor is directly related to its staging, but, every day, the research focuses on the discovery of biomarkers that may help in the determination of the prognosis for this neoplasia. Therefore, this study proposes to evaluate the immunoexpression of important inflammatory markers, as well as nuclear transcription protein and cell proliferation marker, correlating them with prognostic factors in oral carcinomas. A retrospective study was carried out with patients with squamous cell carcinomas in the mouth from 2011 to 2016, in which the medical records were collected for the collection of socio-demographic and clinical-pathological data, as well as the use of the surgical pieces of the same for Immunohistochemistry, through TMA for inflammatory markers (TNF-α and COX-2), transcription (NF-κB) and cell proliferation (Ki67). Immuno-tagging was evaluated qualitatively and quantitatively using ImageJ software, and data were correlated with prognostic factors and patient survival, obtained by the difference between the date of death and the date of initiation of the treatment, expressed in Months. It was observed that the negative and weak TNF-α immunoexpression in primary tumor influenced, improving the survival of patients and that this marker is positively associated with a moderate and intense expression of the other markers studied in both primary tumor and In perilesional tissue and lymph node metastasis. Although COX-2, NF-κB and Ki67 do not show any relationship to improvement or worsening of survival.
