Resumo: Acromegaly is a chronic and rare disease caused by dysfunction of the pituitary gland, which leads to overproduction of growth hormone (GH), associated with increased extremities, multiple organ dysfunctions and physical limitations. Objective: To elaborate, legitimize and validate a manual for people with acromegaly. Material and Method: Descriptive, qualitative and quantitative study. Accomplished in four stages, the first one was made interviews with patients from the acromegaly outpatient clinic of the Sorocaba Hospital Complex, observing their difficulties and physical deficiencies, which allowed to create categories and subcategories. Supported in the previous stage, in the second stage the construction of the first version of the educational manual was carried out. In the third step, the manual was validated with medical judges and physiotherapists, using the five-point Likert scale and analytical tests, measuring reliability and consistency, using the Kappa index and the chi-square test. In the fourth stage, due to the low concordance between judges and, according to what was proposed by the results extracted, a new version of the manual was elaborated, based on the judges' recommendations. Results: In the first step, the categories and subcategories were created from the data collected; the first category was related to the diagnosis with four subcategories, fear of death, anguish, sadness and lack of information; in the second category, difficulties were included in the subcategories, related to osteotic pain, loss of mobility, libido and emotional changes. With the results extracted from the first stage, the first version of the manual was elaborated and, for its validation, it was sent to the judges with the Likert scale, where the Kappa index showed a weak agreement between the groups in the Chi- square the significance was p-value <0.01 evidencing the lack of interdependence between the groups. In view of the results, the second version of the manual was created, adding what was suggested by the judges, and then printed and disseminated in digital media
