Summary: | Morphological, isoenzimatic and molecular markers were used to characterize commercial rice cultivars. The morphological characterization consisted in two experiments, the first one was done in green house, at Federal University of Lavras, when the cultivars were characterized using the descriptors recommended for the protection cultivar law in Brazil. The second experiment was in a seed production field, in EPAMIG, Lavras. In this experiment, contaminants rice seeds were identified for researchers among studied cultivars. In the isoenzimatic characterization, was analyzed isoenzimes from seeds and tissue of rice plantlets, using horizontal gel electrophoresis. This part of the research was done in Embrapa-cpact, Pelotas, RS. The isoenzimatic systems analyzed were esterase, 6-fosfogluconate dehydrogenase, isocitrate desydrogenase and leucine aminopeptidase. For the similarity analysis was used the Jaccard coefficient. The molecular analyses of microsatellites had the objective to identify microsatellites in or around rice genes, to develop primers to the microsatellites selected and to characterize the rice cultivars using the primers developed. Thirteen microsatellites were selected in upstream region of Mads Box genes and 37 microsatellites in ESTs (Expressed Sequence Tags). To search and to select microsatellites were used the databases from rice genome sequencing and used bioinformatics tools available at Plant Research International, Holland, where this research was developed. To analyze the microsatellites was used polyacrilamide gel 6% and the gel was submitted to silver staining. The similarity analysis was done the same way of isoenzyme analysis. The PIC (Polymorphic Informative Content) of microsatellites markers was among 0,05 and 0,955. The conclusions were that the morphological descriptors were useful, but not enough to characterization and discrimination of rice cultivars, the morphological characteristics observed after the reproductive phase are the more suitable to characterization and discrimination of rice cultivars. Isoenzimatic markers allow to characterize rice cultivars and are capable to differentiate all upland rice cultivars studied, excluded Carisma, IAC 202 and Guarani. The esterase isoenzymes are influenced for the presence of fungus in the seeds and the isoenzymes of 6- fosfogluconate dehydrogenase can be altered according to physiologica quality of the seeds. The microsatellites analyzed allows conclude that is possible to find microsatellites in or around rice genes. The analysis of the microsatellites studied allows separate the cultivars according the subspecies, indica or japonica.O interesse e a importância da caracterização de cultivares tem aumentado muito nos últimos anos, sendo os motivos principais, o controle da pureza genética em lotes de sementes e a crescente necessidade do registro e proteção de cultivares. O objetivo deste trabalho foi caracterizar cultivares comerciais de arroz utilizando marcadores morfológicos, isoenzimáticos e moleculares. A caracterização morfológica consistiu de dois experimentos. O primeiro foi conduzido em casa de vegetação, na UFLA, quando foram caracterizadas as cultivares utilizando-se os descritores morfológicos recomendados para o registro e proteção de cultivares. O segundo experimento foi conduzido em campo de produção de sementes da EPAMIG Lavras e consistiu na identificação, por parte de avaliadores, de contaminantes propositalmente distribuídos junto às cultivares. Na caracterização isoenzimática, realizada na EMBRAPA-CPACT, Pelotas, RS, foram analisadas isoenzimas de sementes e folhas de plântulas, utilizando para tal eletroforese horizontal em gel de poliacrilamida. Foram analisadas isoenzimas de esterase, 6-fosfogluconato desidrogenase, isocitrato desidrogenase e leucina aminopeptidase. Foi realizada a análise de agrupamento das cultivares de acordo com a similaridade apresentada, utilizando o coeficiente de Jaccard. A análise molecular de microssatélites teve como objetivos identificar microssatélites em e próximos a genes, desenvolver primers para esses e, com a utilização desses primers, caracterizar as principais cultivares de arroz em uso no Brasil. Foram selecionados treze microssatélites na região anterior aos genes Mads Box, além de 37 microssatélites em ESTs (Expressed Sequence Tags). Foram utilizadas, para a busca e seleção de microssatélites, as bases de dados oriundas do sequenciamento do genoma do arroz. As buscas foram realizadas com o auxílio de ferramentas de bioinformática disponibilizadas pelo Plant Research International, Holanda, onde foi realizada a presente pesquisa. Foi utilizado gel de poliacrilamida a 6% sendo os géis corados com prata. A análise de agrupamento foi realizada de modo similar aos dados oriundos da análise de isoenzimas. Além disso, foi calculado o conteúdo informativo do polimorfismo, tendo sido encontrados valores entre 0,05 a 0,935. Ao final da caracterização dos genótipos, concluiu-se que os descritores morfológicos não foram suficientes para a caracterização e diferenciação das cultivares de arroz estudadas, que as características morfológicas observadas em sementes e plantas após a antese são as mais adequadas para a caracterização e diferenciação de cultivares. A partir dos marcadores isoenzimáticos concluiu-se que a análise de isoenzimas de sementes e folhas de plântulas permite a diferenciação das cultivares, á exceção das cultivares Carisma, IAC 202 e Guarani. Isoenzimas de esterase são influenciadas pela presença de fungos nas sementes e isoenzimas de 6-fosfogluconato desidrogenase podem ser alteradas em função do nível de qualidade fisiológica das sementes.
