This study objective to investigate the profile protein of the seminal plasma in dogs chemically sterilized. Three dogs, pubescent mongrel, were submitted for two semen collections before (D0) and 60 days after chemical castration (D60). Semen samples were analyzed (concentration spermatic, motile, vigor and volume) and centrifuged to obtain seminal plasma. An aliquot containing 400 mg total protein were subjected to 2-D electrophoresis (18% SDS-PAGE, pH 4.7) gels and analyzed by the PD Quest software. The concentration spermatic, motile, viability and ejaculate volume were within normal physiological standards for dogs. Was (220.6 ± 10.97 X 106 sperm / mL, 80%, 3.3 ± 1.53 and 5 mL, respectively). Sixty days after chemical castration all animals were azoospermic but continued to ejaculate the seminal plasma whose volume was 3.67 ± 2.68 mL. The most abundant proteins in the seminal plasma dog are around 12 kDa and pH range from 4.2 to 6.6. These proteins corresponded to 61.68% of total seminal plasma-protein dog, being found 167.3 ± 23.6 spots protein did not differ between D0 and D60. Since the prostate is an androgen-dependent organ responsible for most of the secretion of seminal plasma canine. Oliveira et al (2012) demonstrated that chemical castration has no effect on serum testosterone levels in dogs. However, Wang (2002) reported that after 24 months of intratesticular injection of zinc prostate showed a reduction of 52% of its original size. We conclude that 60 days is not enough to promote changes in the protein electrophoretic pattern in seminal plasma of dogs subjected to chemical castration. |
