Polysaccharides are employed in various industries as thickeners, detergents, laxatives, fiber and food toppings . New research has focused on the anti - mutagenic anti-inflammatory and imunomodulator potential of microbial exopolysaccharides (EPS), as well as in their application in controlled drug delivery systems. Several studies have employed fungal β -glucans and verified high biological activity. The goal of this project is to study the immunomodulatory action of EPS produced by Lasiodiplodia theobromae, evaluate the ability to act as drug carriers of blends formed by exopolysaccharide and latex and also some mechanical properties of these blends. The micro-organism was grown on Vogel medium with sucrose 50 g / L at 28 ° C, 180 rpm for 72 h. The EPS was precipitated with ethanol ( 3:1 v/v), dialyzed and purified by freezing / thawing and ultrafiltration. The yield of EPS in each culture was on average 1.9 g / L. The study of the immunomodulating effects was performed in vitro, only with the fraction of EPS β- glucan ( 1 → 6 ) purified by adding different concentrations of polysaccharide in human U937 macrophages lineage and verifying cell viability and cytokine ( TNF - α and IL – 10 profile ). Concerning the tests of immunomodulation, the EPS purified β- glucan ( 1 → 6 ) has shown to be capable of activate the macrophage response to the release of TNF - α cytokine in the first 24h , decreasing after 48h, without showing response activation of IL - 10 , which suggests a pro-inflammatory and anti- tumor response. The films were evaluated for thickness, moisture loss, water solubility and mechanical strength. The evaluation of the films as a carrier of drugs was performed by the addition of ketoprofen to EPS and latex blend, and monitoring of drug release by spectrophotometry.
