The aim of this study was to evaluate the influence of stages of anestrus and diestrus in oocyte diameter, configuration of chromatin in GV stage and competence of canine oocytes, as well as analyze the kinetics of p34cdc2 kinase activity during in vitro maturation of canine oocytes after 24, 48 and 72 hours of in vitro culture. Ovaries were obtained from bitches after elective ovariohysterectomy (OSH). To evaluate the diameter and configuration of chromatin in GV stage, cumulus-oocyte complexes (COCs) were subjected to 0,2% hyaluronidase solution for removal of cumulus cells and stained with Hoechst. The assessment of nuclear configuration of canine oocytes was analysed after 72 hours in vitro maturation. To perform the ELISA was performed an extract of proteins from oocytes after 24, 48 and 72 hours of in vitro culture. For the mean oocyte diameter, the results showed no difference (p> 0.05) between phases of anestrus and diestrus (77,62 μm x 78,64 μm). Comparing the reproductive phases, higher frequency of oocytes in stages of MI (19,1%) and M-II (14%) was observed in the diestrus phase. However, these differences were not significant (p> 0.05). Four stages of chromatin configuration identified as GV-1, GV-2, GV-3 and GV-4 were classified. The observed changes in chromatin configuration been characterized as a transition dispersed chromatin (GV-1, GV-2) for partially condensing (GV-3) until it reaches a fully condensed phase (GV-4). The results showed difference (p<0.05) between phases of anestrus and diestrus, with a higher proportion of GV-1 and GV-2 during anestrus and GV-3 and GV-4 during diestrus. The p34cdc2 kinase activity was shown to be time-dependent, reaching a peak at 48h of IVM (p<0,001). After 72h, the activity showed a decrease. Thus, identification of factors related to the resumption and progression of meiosis in vivo is critical for making appropriate adjustments and improvement of culture conditions |
