Produção de filmes de Ecovio®/quitosana/poli (óxido de etileno) por eletrofiação para liberação de ibuprofeno

Resumo: | Ibuprofen is a drug that has anti-inflammatory, analgesic and antipyretic effects. As it is a drug widely used in the treatment of pain and fever, there is great interest in its study due to the side effects that can be generated by daily oral use. Thus, this work aims at the production of polymeric films of ecovio® (polymeric blend composed of polylactic acid and poly(butylene-adipate-co-terephthalate)/chitosan/poly(ethylene oxide)) for transdermal delivery of ibuprofen, obtained by electrospinning. Initially, a polymer solution consisting of chitosan and poly (ethylene oxide) (90/10% - m/v) in a solution of water and acetic acid (10/90% - v/v) was prepared. This solution was electrospinned, with a 20 cm distance between the needle and the collection plate, 0.5 mL/h of flow and 18 kV of applied potential difference. Then, ecovio® (15% - m/v) was associated with the drug ibuprofen (30% - m/v) in a solution of chloroform and dimethylformamide (85/15% - v/v) and this solution was electrotrophied on the film composed of chitosan and poly(ethylene oxide) with 12 cm of distance between the needle and the collecting plate, 1.0 mL/h of flow and 18 kV of applied potential difference. Four more films were produced, two by electrospinning (ecovio®/chitosan/PEO and ecovio® + ibuprofen) and the other two films (ecovio® + ibuprofen and ecovio®/chitosan/PEO + ibuprofen) were manufactured by casting. The morphological characterizations of these films were performed by SEM and showed an increase in the number of beads and fiber discontinuity. Furthermore, physicochemical characterizations were performed using TGA, DSC and x-rays, which showed the incorporation of the drug in the films. The release study was performed under conditions that simulate the condition of the skin, according to pharmacopoeial parameters. The method used to quantify ibuprofen during the release study showed selectivity and linearity, as the interference percentages of the absorbances of the ecovio®/chitosan/PEO components exhibited results below 2% compared to the standard solution, and the Pearson's correlation coefficient obtained was equal to 0.9999. Therefore, the method is selective and linear for the quantification of ibuprofen in the range of 0.0034 mg/mL to 0.0204 mg/mL. The electrospinned film containing ecovio®/chitosan/PEO + ibuprofen released, in 30 minutes, approximately 95% of the active substance, being 100% released in 90 minutes, that is, the film presented a rapid release profile. Thus, the ecovio®/chitosan/poly (ethylene oxide) film incorporated with ibuprofen showed promise for use in transdermal delivery. On the other hand, the electrotrophied film of ecovio® + ibuprofen and the films produced by casting showed release results in the range of 60-80% of the drug, which indicates that in 90 minutes there was no complete release of the drug available. |
