Resumo: | Introduction: The resulting fibrosis in the healing process can be steeper than usual forming hyperproliferative skin scarring such as hypertrophic scar and keloid, which latter is unique to humans. Both scars have more nerve fiber endings, compared to normal skin, suggesting that neuropeptides and neurotrophins were involved in generating, maintaining and postoperative recurrence of these scars. Objective: To evaluate the expression of 84 genes related to neurotrophins and receptors in keloid. Methods: Were collected twelve (12) samples of keloid in keloid group and twelve (12) normal skin samples in the control group. Total RNA was purified with Qiagen RNeasy Minikit DNAse and the quantity and quality of the extracted RNA were assessed by spectrophotometry. The mRNA was used for cDNA synthesis. The reverse transcription reactions were carried out using the First Strand Kit RT2 Superarray Bioscience, according to the manufacturer's protocol and RT-qPCR was performed using the PCR RT2 Profiler® Superarray Bioscience array 84 where relevant human genes involved were analyzed the neurotrophin synthesis and their receptors - Human neurotrophins & receptors PCR Array (PAHS-031Z). Results: Twelve (12/84 or 14%) genes - HSPB1, GRPR, CRHR2, NPY2R, 1L6ST, NGF, MT3, BCL2, CCKAR, PSPN, GalR2 and FRS3 were differentially expressed in keloid. Conclusion: The differentially expressed genes, 100% hypo-expressed in keloid compared to normal skin, and priority genes are HSPB1, GRPR, CRHR2, NGF, MT3 and BCL2. |
