Resumo: Introduction: The cultive of periodontal ligament fibroblasts associated with simulation of pressure or tension on the cell allows analysis in its morphology and form, identify proteins and molecules involved in this process. The system formed by the protein Nuclear Factor Activator Receptor ?B (RANK) \ Nuclear factor Activator Receptor ?B Ligand (RANKL) and osteoprotegerin (OPG) are directly linked to the regulation of osteoclast and osteoclastogenesis, formation and / or bone resorption. Objective: Evaluate the fibroblasts of RANK \ RANKL and OPG in adult fibroblasts of human periodontal ligament under continuous pressure. Methods: The tissue was collected from periodontal ligament of 40 third molars of adult patients who were grown in culture flasks until the sixth passage. The cells were placed on plates with 6 wells with a 3 mm diameter which were subjected to the continuous pressure (3 to 4 g / mm2) for 6 hours in monolayers. From the samples, the mRNA was extracted and evaluated by method Polymerase Chain Reaction Real-Time (qPCR).Results: In the continuous pressure model by using the fibroblasts of the periodontal ligament, in all the groups there was no expression of RANK \ RANKL, and all groups expressed OPG, and was significantly higher in the experimental group 2 (EG2) compared to control (CG) and experimental group 1 (EG1) (P = 0.005). Conclusion: The adult fibroblast from human periodontal ligament when under continuous pressure not expressed mRNA RANK \ RANKL and osteoprotegerin mRNA was expressed.
