Relação entre o perfil metabolômico do secretoma embrionário e seu desenvolvimento in vitro

Resumo: | The need for a noninvasive, reliable and rapid strategy for embryo evaluation has promoted metabolomic studies in the area of assisted human reproduction. The embryo physiology reveals more about the embryo than can be determined by morphology alone, therefore, we believe that the preimplantation embryo metabolism shows characteristics that may be associated with embryonic development potential. In this context, we hypothesized that levels of culture medium metabolites reflect the embryonic development potential. Objectives: Evaluating the embryonic secretome metabolomic profile and examine its relation with (i) morphological quality in vitro culture D3 and (ii) its ability to reach the blastocyst stage. Method: The metabolomic analysis of the embryonic secretoma in the D3 culture medium of human embryos cultured in vitro was performed. For (i) evaluation of the morphological quality to the D3, the samples were grouped according to the embryo development, in High Quality or Low Quality. For (ii) assessment of the competence to reach the blastocyst stage, the samples were grouped according to the evolution to D5 \ D6 in Blastocyst or not Blastocyst. The samples were analyzed using a quantitative and qualitative approach using the AbsoluteIDQ® p180 Kit (Biocrates Life Science, AG, Austria). Metabolites were compared using the MannWhitney U test. Results: The analysis of the profile of the embryonic secretome in the culture medium showed differences between the evaluated groups. Statistical analysis revealed 25 metabolites with statistically different concentrations between embryos that showed high or low quality in D3 and 29 metabolites with different concentrations when evaluated the embryonic competence to reach the stage of blastocyst. Conclusion: the metabolites, products of the embryonic secretome in the culture medium of the third day of in vitro development, reflect embryonic morphological quality in D3 and the competence to reach or not to blastocyst stage. |
