Resumo: | Aim: To evaluate the possible protective effect of the decoupling of oxidative phosphorylation and concomitant stimulation of glycolysis during cryopreservation of human spermatozoa. Methods: Each semen collected through the spermogram was divided into two fractions that received the following treatment: (1) simultaneous treatment of oxidative phosphorylation and stimulation of glycolysis (treated group); The aliquots were cryopreserved and analyzed, before and after this process, through evaluations of oxidative status, structural integrity and sperm motility. The obtained data were evaluated and the level of significance used to reject H0 (null hypothesis) was 5%. Results: The aliquots received from the Treated Group showed more cells with intact acrosomes, less DNA fragmentation and better mitochondrial activity compared to the Control Group. Conclusions: From the results obtained, we can conclude that the treatment had a protective effect on human spermatozoa during cryopreservation: (1) preventing cryoinjury, such as high DNA fragmentation and decreased acrosome integrity; (2) maintaining the integrity and activity of mitochondria. |
