Estratégias analíticas para a obtenção de marcadores, validação e padronização de extratos de Gomphrena vaga Mart (Amaranthaceae)

Resumo: The species Gomphrena vaga Mart. presents an irregular distribution throughout the Brazilian territory, its name being a reference to such characteristic. It is part of the vegetation of forests and cerradões in the southeast and center-west regions. This species is popularly used on the form of infusions (roots and / or aerial parts) as analgesic, tonic and for the treatment of infections of the respiratory tract. This work performed the development of analytical methodologies for the identification, fractionation, isolation and purification of the major constituents of extracts of the species using high performance liquid chromatography (HPLC) at analytical and semipreparative scales from the ethanolic root extract. The major constituents were identified as the ecdysteroids 20-hydroxyecdysone and 1α, 20R-dihydroxyecdysone. After the structural determination of these chemical markers, a CLAE method with diode array detection (DAD) was developed and validated according to ICH and ANVISA standards, for the quantification of these in aqueous and hydroalcoholic extracts of the stems and roots of the species. Based on the validated method, the best extraction conditions for the roots and stems were evaluated in order to obtain the best yield of these two markers, extractor solvent (infusion with water and maceration with hydroethanolic solutions), vegetal drug-extractor solvent ratio and temperature, allowing the obtaining of standardized extracts in 20-hydroxyecdysone and 1α,20R-dihydroxyecdysone with higher efficiency in the conditions ethanol 60% in drug-solvent extraction ratio of 1:10 and infusion (aqueous) in the drug-solvent extractor ratio of 1:5, respectively.
