Resumo: Dihydromyricetin (DMY) is a flavonoid, which has several pharmacological properties with an emphasis on antioxidant, antitumor, photoprotective activity among other beneficial biological effects. However, DMY has low aqueous solubility and consequently, low bioavailability. The objective of this work was to evaluate the safety profile and photoprotector efficacy of hydrogels containing cationic nanocapsules dihydromricricetin (NC-DMY). Results showed no cytotoxicity and genotoxicity in peripheral blood mononuclear cells when exposed to NC-DMY and free-form DMY (DMY-F) suspensions, however, concentration-dependent DNA damage was observed for Eudragit RS100® nanocapsule suspensions. (NC-E). NCDMY and DMY-F showed a high antioxidant capacity, being these dose-dependent demonstrating the permanence of the antioxidant effect in nanoencapsulation. Nanocapsule suspensions, when inserted into hydrogels retained physicochemical characteristics, however, improved stability when exposed to storage at 4 ± 2°C. The highest concentration of the drug DMY remained in the stratum corneum was found in the evaluation ex-vivo skin permeation and there was no hypodermis permeation for the hydrogels containing NC-DMY and DMY-F. Regarding safety, all formulations are safe for topical application to 3T3 cell lines, human fibroblasts and HaCaT. Hydrogels containing NC-DMY showed photoprotection against DNA damage caused by UVB radiation, with Solar Protection Factor (SPF-DNA) close to 50, and 99.9% protection against induction of DNA damage, proving to be a formulation promising with photoprotective and antioxidant potential, being able to eliminate free radicals formed by solar radiation.
