Estudo fitoquímico, controle de qualidade e desenvolvimento de formulação fitoterápica do óleo resina de Copaifera officinalis (copaíba) para terapia adjuvante de pacientes oncológicos.

Resumo: | Copaibas are tree species belonging to the Fabaceae family and the genus Copaifera, native to tropical regions. An oil exudes from the trunk of copaib trees, consisting of a resinous fraction rich in diterpenic acids diluted in a volatile fraction, rich in sesquiterpenes. This oil is used in traditional medicine for its healing, anti-inflammatory and antimicrobial properties, already proven in in vitro and in vivo studies. The objective of the study was to carry out a phytochemical evaluation for the characterization and physical-chemical quality control of the resin oil of Copaifera officinalis L. and to develop an innovative formulation. In the qualitative phytochemical study, 64 substances were identified in the resin oil by gas chromatography coupled to the mass spectrum (GC-MS), the majority being the sesquiterpenes copaene, β-caryophyllene, humulene and the diterpenes kolanelool and hardwickiic acid. For qualitative/quantitative analysis, an analytical method by GC-MS was developed and validated to quantify the selected chemical marker, β-caryophyllene. The method was validated and proved adequate, showing selectivity, linearity (r2 ≥ 0.98.), accuracy (110-125%), precision (DPR ≤ 5%), being reproducible and robust to small variations in analytical conditions. In addition, a qualitative method of analysis of C. officinalis resin oil by thin layer chromatography was developed. For the quality control of the oil, analyzes of the physical chemical parameters of density (1.0170 g/mL) and viscosity (25.6498cp) were carried out. The developed pharmaceutical foam of copaiba oil is biphasic and stable, with a foaming capacity of 1.53; 53% foamability; good spreadability without touch (19 cm2), with pH suitable for topical application (5.2). The study demonstrates the authenticity and purity of copaiba resin oil; proposes qualitative and quantitative analysis methods and quality parameters for the development of a pharmacopoeial monograph for the species C. officinalis and opens up perspectives for preclinical and clinical confirmation of the pharmacological activities of copaiba resin oil and the developed formulation. |
