Desenvolvimento de testes de PCR para BoHV-5 e sua aplicação no diagnóstico de casos clínicos.

Resumo: The bovine herpesvirus are economically important pathogens causing a variety of clinical syndromes. Neurological manifestations only currently assigned to bovine herpevírus 5 (BoHV-5). The genome of BoHV-5 and BoHV-1 have 85% similarity, making it difficult to differentiate by traditional diagnostic techniques. The development of tests for the detection of BoHV-5 is important for the differential diagnosis of other diseases that induce neurological symptoms in cattle, such as rabies. We developed a PCR test for the US4 region of BoHV-5, using field samples EVI-88 and A663 (BoHV-5) as standard. The test was standardized for use in cell culture supernatants or after DNA extraction. The test specificity was tested against samples of human and animal herpesvirus, and also by sequencing the DNA fragment obtained from the EVI-88. The sensitivity was shown 10 to 3.25 TCID50 / 50l. or 0.0006 TCID50 / 50l. for cell culture supernatant and 10 to 1.25 TCID50 / or 0.06 TCID50 50l / 50l for DNA extraction. We tested 20 clinical samples of paraffin central nervous system of cattle with encephalitis with a positivity of 35%. The developed PCR is presented as an alternative rapid, sensitive and specific for detection of BoHV-5 in clinical samples
