Resumo: | The endogenous bleaching techniques present varying degrees of toxicity and can often lead to development of cervical inflammatory resorption. In this study, it was evaluated the immune response to the application of bleaching agents.. Additionally it was studied the same parameters in regards to repairing agents used in the treatment of cervical resorption. It was inoculated the bleaching agents, sodium perborate mixed with hydrogen peroxide [20 volumes] (PS+H2O2 20V) and carbamide peroxide [37%] (PC) in the skullcap of mice. After 7 and 14 days, calvaria have been extracted: tartrate-resistant acid phosphatase (TRAP) expression was defined by histology and cytokines were assayed by real time PCR. Using the same animal model, it was evaluated the effects of repairing agents MTA, Emdogain® and Geristore® in regards to cytokine expressions, at 14 and 21 days after surgical procedures. The results showed that PS+H2O2 20V and PC induced the expression of TRAP in both periods. The gene expression of RANKL, TNF-, IL-6 and IL-4 was induced by PC at 7 and 14 days. On the 14th day this agent induced higher expression of IFN- than PS+H2O2 20V. The gene expression of RANKL and TNF- were induced by PS+H2O2 20V on the 7th and 14th day. On the other hand, this agent induced low expression of IL-6 and IFN- at 14 days. Concerning to repairing agents, it was observed Emdogain® suppressed the gene expression of RANKL, RANK, OPG, IL-6, IFN-, IL-10, IL-4 and TGF-, on the 14th day. MTA did not induce or suppress the expression of cytokines on the 14th day. Geristore® did not alter or suppress the gene expression of all mediators, in both periods, except for TNF- (14 and 21 days), IL-6 and IL-4 on the 14th day. The induction of RANKL, RANK, OPG, IL-6, IFN-, IL-10 and IL-4 was observed in response to Emdogain®, on the 21th day. However, it was significantly lower than that observed to MTA. In contrast, in this same period, the MTA induced expression of TGF-, slightly lower than the Emdogain®, and it did not induce the expression of TNF- in any period. We can conclude the model used in this study, a pioneer in evaluating the effects of bleaching agents and repairers on the activity of bone cells, was able to mimic the action of these agents, allowing longitudinal evaluation of the mediators involved in the phenomena of bone resorption and repair. |
