Resumo: | The spleen is an organ of great importance, especially in the defense mechanism of the body, and its absence may lead to fulminant sepsis. Total splenectomy therefore should be avoided and research should be conducted to increase the options in the maintenance of splenic tissue. Objective: Verify if there is difference between the phagocytic function of the lower splenic pole remaining of subtotal splenectomy and autogenous splenic implants. Methods: 36 male Wistar rats , weight 364 ± 60 g , underwent subtotal splenectomy preserving the lower pole and autogenous splenic implant into the greater omentum . These animals were divided into seven groups according to the time of euthanasia : euthanasia with a G1 - day postoperative period ( n = 5 ) , G7 - seven days postoperatively ( n = 6 ) , G30 - 30 days PO ( n = 5 ) , G60 - 60 days postoperatively ( n = 5 ) , G90 - with 90 days postoperatively ( n = 5 ) , G120 - 120 days postoperatively ( n = 5 ) , G150 - 150 postoperative days ( n = 5 ) . At the time of euthanasia the autogenous splenic implants and the lower pole were weighed. The feasibility was assessed macroscopically. The phagocytic function was assessed by splenic uptake of radioisotope -labeled colloid and macrophages counted by the Cellprofile software program. Results: The viability of the autogenous splenic implants and inferior pole of the spleen was found in 33 ( 94.3 % ) animals , with no difference between them . The weight of the implants was higher than the lower pole in animals in groups G1 , G7 , G30 , G60 and G120 . The phagocytic function of the implants by radioisotope uptake was higher than the lower pole in G7 and G120 groups and did not differ in the other groups . The number of macrophages was higher in G1 , G60 , G90 and G120 and did not differ in the other groups . Conclusion : Until the 16th week the phagocytic activity was more pronounced in autogenous splenic implants compared with the lower splenic pole, but became similar thereafter. |
