Resumo: | Glycation is a non-enzymatic reaction between reducing sugars and amino group of biomolecules, forming advanced glycation end-products (AGEs), which are associated with various pathological conditions such as atherosclerosis, osteoporosis, asthma, arthritis, sarcopenia, Alzheimer's disease, cancer and various diabetic complications. Due to their biological effects, the search for new molecules that inhibit the glycation and the formation of AGEs is of great importance. The present work aims at the synthesis of new derivatives of lipoic acid with potential antiglycation activity. First, the studies carried out for the synthesis of new derivatives of lipoic acid containing the amidine or aminoguanidine group in their structure are described, with the use of different linkers as a strategy to join two subunits. Subsequently, new derivatives of lipoic acid (ester or amide type) were prepared from natural or synthetic products with potential antiglycation activity. Preliminary studies of the anti-glycation activity of the new derivatives were carried out, with three compounds shown to be active. |
