Resumo: The objective of this research was to estimate the acute toxicity, subchronic and the pharmacological effects of the ethanol extract of the stem bark of guatambu (Aspidosperma subincanum) in rodents. To carry out the acute toxicity it was used albino female mice (Mus musculus) that received a single oral dose of 300 mg/kg and 2000 mg/kg of the extract. As for the subchronic toxicity test were evaluated mice (Mus musculus) males and females that received the oral dose of 75 mg/kg, 150 mg/kg and 300 mg/kg of the extract daily for 28 days. To achieve the pharmacological tests, it was used a dose of 125 mg/kg, 250 mg/kg and 500 mg/kg orally in male mice. The animals showed some signs of neurotoxicity such as tremors, changes in the gait and convulsions. Such signals have the intensity proportional to the concentration of the extract, and were lethal in the dose of 2000 mg/kg.It was observed that in acute or subchronic conditions, depending on the dose, the timing and frequency of exposure and routes of administration, the ethanol extract of Aspidosperma subincanum should be considered a toxic agent.
