Correlação da expressão da IL-5 e do estado de ativaçãodo eosinófilo com a eosinofilia tecidual associada ao tumor (TATE) no carcinoma de células escamosas bucal

Resumo: Tumor associated tissue eosinophilia, or TATE, is characterized by infiltrating eosinophils in neoplasias unassociated with areas of necrosis or ulceration. Eosinophils are commonly identified in oral squamous cell carcinomas (OSCC) and may influence patient s prognosis participating in the antitumor immunological reactions. The activation of eosinophils which is characterized by cytotoxic protein secretion is just as important as the number of infiltrating cells. The present study evaluated 27 cases of OSCC in relation to eosinophilia and to the activation status of eosinophils. We also evaluated the existence of a molecular microenvironment usually considered favorable to eosinophilia, being characterized by the presence of interleukin-5 (IL-5), the eosinophil differentiation factor. Il-5 also indicates a Th2-like inflammatory reaction. The quantification of eosinophilia was preformed on Sirius red stained sections. The activation status of eosinophils was accomplished by immunostaining with EG1 and EG2 monoclonal antibodies. EG1 identifies the eosinophil cationic protein (ECP) in its storage and secreted forms while EG2 identifies ECP and eosinophil derived neurotoxin (EDN) in its secreted forms. The molecular microenvironment was also analyzed by immunohistochemistry technique using anti-h-IL5 polyclonal antibody for IL-5 identification. The results showed statistically significant correlation between eosinophilia with EG1 and EG2. However, there was no correlation between eosinophilia and anti-hIL5. The results suggest that in OSCC the infiltrating eosinophils are activated and that other factors besides IL-5 may be responsible for its presence and that these cells are not necessarily restricted to Th2- like reactions
