Resumo: Squamous cell carcinoma of the head and neck is the sixth most common type of cancer worldwide. Several genes and signaling molecules are involved in the development of oral squamous cell carcinoma. The study aimed to analyze the potential applicability of a panel of tumor biomarkers as indicators of tumor progression and prognosis in oral squamous cell carcinoma. The expression profile of the proteins survivin, CDKN2A, EGFR, PLK1, p63, p40, cyclin D1, BCL2 and CDK1 of 109 paraffinized tumor tissue samples for immunohistochemical analysis and 40 fresh tissue samples for the execution of the technique were evaluated. real- time polymerase chain reaction after reverse transcription. Survivin protein overexpression in tumor segments (p<0.001) and strong correlation with the expression of PLK1 (r=0.742, p=≤0.001) and cyclin D1 (r=0.669, p<0.001) proteins. Validating these results, the survivin coding gene showed overexpression in 53.12% of frozen tumor tissues and a strong correlation as to expression with the CDK1 gene (r=0.8573, p<0.0001), when evaluated by the technique of real-time polymerase chain reaction after reverse transcription, reinforcing its potential biological role for the stabilization of survivin in oral squamous cell carcinoma. Survivin stood out as a promising biomarker of progression from epithelial tissue to dysplastic epithelium and OSCC, due to its limited expression to tumor tissue and its correlation with key proteins involved in cell cycle transition, such as PLK1, p63, p40, therefore, strong candidates for the construction of a panel of biomarkers with potential applicability in clinical routine.
