Efeito nefroprotetor do extrato alcoólico de Lippia sidoides em modelos de lesão renal aguda por isquemia/reperfusão

Resumo: Ischemia/reperfusion (I/R) in kidney is commonly related to acute kidney injury (AKI), essentially through oxidative stress. Lippia sinoides is an aromatic shrub that grows wild in the northeastern region of Brazil that has anti-inflammatory and anti-oxidative properties. Lippia sinoides (LS) protection in renal I/R injury was investigated. Male Wistar rats underwent unilateral nephrectomy and contralateral renal I/R (60min). Rats were divided into four groups: (1) Sham group, (2) LS group (sham-operated rats treated with LS), (3) IR group (rats submitted to ischemia), (4) IR-LS (rats treated with Lippia sinoides before ischemia). At 48h after reperfusion, renal function and oxidative stress were assessed and kidneys were removed for analysis to evaluate the protective effect of LS on renal damage induced by I/R. It was also evaluated the protective effect of LS on the renal cell lineage against I/R damage. I/R caused alterations in plasmatic creatinine, urea, creatinine clearance and microalbuminuria. Treatment with Lippia sinoides ameliorated all of these parameters. In addition, KIM-1, N-GAL and Nephrin levels enhanced by I/R was also diminished in groups treated with LS. The histological examination showed that LS attenuated the morphological changes caused by I/R, markedly vascular congestion and intratubular deposits of proteinaceous material. Additionally, LS was able to reduce the changes observed in TBARS and GSH levels in kidney tissue. In in vitro assay, LS was capable to partially protect the cell lineage against cell damage induced by I/R. Lippia sinoides has a nephroprotective effect in kidney against AKI induced by I/R, with antioxidant effect. Moreover, this result seems to be associated to a direct protective effect on tubular epithelia.
