Atividade antimicobacteriana do óleo essencial da casca de Cinnamomum zeylanicum e do trans-cinamaldeído e suas associações com rifampicina e isoniazida

Resumo: The World Health Organization (WHO) has estimated that 10.4 million new cases of tuberculosis (TB) occurred worldwide in 2015, with a steady increase in resistance cases. Considering TB control difficulties, it is necessary to discover new drugs, especially for resistant strains, and it is believed that medicinal plants, through their essential oils (EOs), are one of the alternatives for bioprospecting active substances, such as the EO extracted from the bark of Cinnamomum zeylanicum (EOCzey), popularly known as cinnamon, and trans-cinnamaldehyde (TCin), its compound, which has several therapeutic properties, such as antimicrobial, anti-inflammatory, and hypoglycemic action, among others. In this context, the present study evaluated the antimycobacterial action of EOCzey and TCin on sensitive and resistant clinical isolates (CI) of M. tuberculosis, as well as the effects of the associations between EOCzey and TCin and the drugs rifampicin (RIF) and isoniazid (INH) on the standard M. tuberculosis H37Rv strain. The Resazurin Microtiter Assay (REMA) technique was used to determine the minimum inhibitory concentration (MIC) of the compounds tested on nine M. tuberculosis CIs. The effects of the EOCzey / RIF, EOCzey / INH, TCin / RIF and TCin / INH associations on the standard M. tuberculosis H37Rv strain were evaluated according to the checkerboard method, determining the Fraction Inhibitory Concentration Index (FICi), in which the synergistic effect was observed with FICi values ≤ 0.5, additive effect with FICi >0.5 and ≤ 1 and antagonism with FICi >1.0. It was observed that EOCzey and TCin were able to inhibit the growth of all nine sensitive and resistant CIs with MIC values ranging from 9.75 μg / mL to 39 μg / mL. All four associations tested resulted in synergistic and additive effects, with no antagonistic effect. Both EOCzey / RIF and EOCzey / INH associations showed FICi values of 0.375 and 0.5, whereas the TCin / RIF and TCin / INH associations showed FICi values of 0.31, 0.375 and 0.5.
