Efeito da composição bioquímica do plasma seminal de caprinos sobre os espermatozoides da cauda do epidídimo e ejaculado resfriados a 5º C

Resumo: The present study was carried out to evaluate the effect of biochemical composition of seminal plasma (SP) on spermatozoa from caudal epididymis and ejaculated goat sperm, diluted in egg yolk-citrate (EYC) and egg yolk-tris (EYT). The ejaculates of 8 animals were collected in the morning and fresh semen was divided in order to obtain 4 treatments: EYC (washed ad no washed), EYT (washed ad no washed). On the same day, in the afternoon, the animals were orchidectomized to obtain spermatozoa from caudal epididymis, of which four aliquots of 100 L were removed. Two of them were added 120 L of autologous seminal plasma, consisted in other 4 treatments: EYC (spermatozoa from caudal epididymis with and without seminal plasma) e EYT (spermatozoa from caudal epididymis with and without seminal plasma). The seminal plasma was evaluated for biochemical composition. Samples of 8 treatments were analyzed for vigor, motility and sperm morphology, at 0 hour (fresh) and after 2 and 24 hours in storage at 5ºC. Data were analyzed with the statistic software SAS@. The following results were found. In grouping by the concentration of fructose and chlorides was observed a significant effect (p <0.05) independent of diluent and treatment, for ejaculated spermatozoa with minor defects and normal. From the potassium concentration was showed that only the vigor suffered group effect (p <0.05). In the spermatozoa from caudal epididymis, fructose was a significant effect for group interaction, treatment and time on motility, for the vigor there was a significant interaction effect between treatment and time and group effect, in the fructose and potassium concentrations, respectively. For the chloride concentration was observed a significant interaction between group and treatment on the occurrence of major defects, (p <0.05). After 24 hours of storage, the best values were observed in sperm without seminal plasma from caudal epididymis, however in the presence of seminal plasma, sperm from the ejaculate withstood better conservation (p <0.05). The addition of seminal plasma reduced seminal parameters of the sperm from caudal epididymis. The biochemical composition of seminal plasma of goats raised in northeastern Brazil interferes directly on the quality of cooled semen.
