Identidade genética em locos de SSR e EST-SSR na cultivar RB92579 de cana-de-açúcar (Saccharum spp.) cultivada nos estados do Paraná e Mato Grosso do Sul

Resumo: | The objective of this study was to evaluate 92 samples of sugarcane about their genetic uniformity in four loci SSR and eight loci EST-SSR in the variety RB92579, collected in Paraná and Mato Grosso do Sul state. These six loci (EstC80, Smc226CG, EstC69, EstB58, mSSCIR52 and EstC45) presented polymorphism while the other six loci (Smc2007, EstC119, EstC84, Smc477CG, EstC113 and EstB149) were monomorphic; therefore the proportion of polymorphic loci was 50%. Monomorphic loci can be recommended for fingerprint cultivar RB92579 as an auxiliary tool to the traditional varietal identification processes. For the 12 loci, 25 alleles were found with an average of 2.08 alleles per locus analyzed. The number of alleles at single repeated sequences which are expressed (EST-SSR) was lower (2.0 alleles per locus) than the number of alleles at loci single repeated sequences which are not expressed (2.25 alleles per locus). The expected heterozygosity values (He) and Shannon information index (I) were slightly higher for samples of RB92579 grown in Paraná (He = 0.098 and I = 0.1455), than the samples grown in Mato Grosso do Sul (He = 0.086 and I = 0.1327). The molecular variance analysis showed that the highest percentage of genetic variation (96%) is within the samples of the two populations, while among these samples of Mandaguaçu-PR and Angelica-MS the molecular variance was only 4%. The analysis of the genetic structure showed that the variety RB92579 is formed by a set of plants containing ancestral alleles of three groups (K = 3) in the analyzed loci. It was observed little mixing of alleles, 80.44% of the evaluated plants have more than 90% of only one of the three ancestral genomes; only 19.56% of the plants have mixing in different proportions two or three ancestral genomes. The EstC69 loci, which encodes the cellulose synthase-6 enzyme, has two alleles. The Shannon information index (I) for this locus was 0.5850, the expected heterozygosity (He) was 0.3958, and observed heterozygosity (Ho) was 0.5435. The FIS for this locus was -0.4145, indicating an excess of heterozygotes. The genetic identity between samples of sugarcane of the two collection places calculated for the 12 loci was 0.997, indicating high genetic similarity, so it can be inferred that the procedures carried out by the sugar industry may be the same for RB92579 variety of samples from Mandaguaçu-PR and Angelica-MS. |
